A study of pearl oysters Pinctada margaritifera growth and mortality was done on a monthly basis between June 1990 and September 1991 in Takapoto atoll (Tuamotu archipelago). Meanwhile, sampling parameters representative of bivalves food showed a remarquable spatial stability whithin the lagoon. The amplitude of temporal variability varied in the order of some days. Carrying capacity appears low : energetic content of particulate material in waters is about 2 joules/liter, ten times lower than in temperate areas. On the opposite, seasonal effect is reduced and particulate food is always available. Results of research concerning the food of the pearl oyster Pinctada mai margaritifera are presented, by taking Takapoto atoll (Tuamotu archipelago, French Polynesia) as a study site. Monthly sampling of several parameters representative of bivalve environment and food (water temperature, salinity, mineral and organic matter, proteins, lipids and carbohydrates, chlorophyll a and phaeopigments) confirm and increase knowledge of the Takapoto lagoon hydrobiology. The variability of these parameters was analysed. Takapoto lagoon showed a remarkable spatial stability. Temporal variations showed a low seasonal trend. Exploitation of the pearl oyster Pinetada margaritifera, which has developed in French Polynesia over the last twenty years, is based mainly on the collection of natural spat. Most bivalves species of the genus Pinctada are well known throughout the world for production of white or black pearls of high commercial value. For cultured pearl production, a mantle allograft from a donor is implanted into the gonad of a recipient oyster, together with a small inorganic bead. Because of the dedifferentiation of cells during the first steps of the host oyster's immunological reaction, so far the fate of the graft and its exact role in the process of pearl formation could not be determined via classical histological methods. In order to study further the genetic structure of the pearl oyster Pinctada margaritifera in French Polynesia with a special consideration for the sampling scale, we analyzed or re-analyzed sets of data based on nuclear DNA markers obtained at different spatial scales. At a large scale (several 1,000 km), the remote Marquesas Islands were confirmed to be significantly differentiated from Tuamotu-Gambier and Society archipelagos, with a marked difference however for the two main islands that are different from each other. Preliminary experiments allowed defining basic requirements of Pinctada margaritifera for somatic and gonadic growth in controlled conditions. This study was continued by observation of the gametogenesis at a cellular level. In April/May 2008, 125 oysters were conditioned for 30 days in 25 tubular tanks (30 L). Pearl oyster grafting is a complex surgical operation that should lead to pearl formation after approximately eighteen months. Although this technique has been used for many years in French Polynesia, the grafting process is still not standardised. While studies have been carried out in order to improve graft performance and yield, these remain highly variable due to post-grafting mortality, nucleus rejection and unreliable pearl quality, all of which constrain pearl farm profitability. Cryopreservation is a valuable tool for genetic improvement programs. Several bivalve mollusc species have already been the subject of such programs and the Tahitian black pearl oyster industry is now planning the development of selective breeding for desirable traits in Pinctada margaritifera. The ability to cryopreserve spermatozoa would, therefore, offer significant benefits to the cultured black pearl industry. Spermatozoa were cryopreserved with CPA 0.7 M trehalose in 0.8 M Me2SO and a two-step freezing process was used: straws were first maintained in nitrogen vapour for 10 minutes, then directly plunged into liquid nitrogen and stored for one week before use.
