The population of Pinctada margaritifera and Pinctada maculata lay eggs throughout the year. Temperature variations induce huge but asynchronous laying. Preferential periods of gonad maturity correspond to the season changings in both species. However. some differences between the two specics are noticed. Both species are protandric hermaphrodites. Female condition is unstable: a reversion to the male condition may be observed and is probably linked with stress. A study of pearl oysters Pinctada margaritifera growth and mortality was done on a monthly basis between June 1990 and September 1991 in Takapoto atoll (Tuamotu archipelago). Meanwhile, sampling parameters representative of bivalves food showed a remarquable spatial stability whithin the lagoon. The amplitude of temporal variability varied in the order of some days. Carrying capacity appears low : energetic content of particulate material in waters is about 2 joules/liter, ten times lower than in temperate areas. On the opposite, seasonal effect is reduced and particulate food is always available. L'huître perlière, Pinctada margaritifera (Linné, 1758) var. cumingi (Reeve), productrice de perles noires, fait l' objet d'une aquaculture florissante en Polynésie Française. Afin de pérenniser cette activité, un programme de recherche (le PGRN) a été lancé, en 1990, sur l' atoll de Takapoto. Virus-like particles in pearl oyster Pinctada margaritifera ArchiMer

Virus-like particles were detected in granulomas associated with focal necrosis within the adductor muscle of the pearl oyster Pinctada margaritifera from French Polynesia. Particulate matter in Takapoto lagoon waters: potential food for cultivated pearl oysters ArchiMer

Results of research concerning the food of the pearl oyster Pinctada mai margaritifera are presented, by taking Takapoto atoll (Tuamotu archipelago, French Polynesia) as a study site. Monthly sampling of several parameters representative of bivalve environment and food (water temperature, salinity, mineral and organic matter, proteins, lipids and carbohydrates, chlorophyll a and phaeopigments) confirm and increase knowledge of the Takapoto lagoon hydrobiology. The variability of these parameters was analysed. Takapoto lagoon showed a remarkable spatial stability. Temporal variations showed a low seasonal trend. Developmental aspects of biomineralisation in the Polynesian pearl oyster Pinctada margaritifera var. cumingii ArchiMer

The shell biomineralisation with special reference to the nacreous region is observed during the development of the Polynesian pearl oyster. Ultrastructural changes were studied and timed for the first time from planktonic larval shells to two-year-old adult shells. During the first two weeks following fertilization, the prodissoconch-I shell structure is undifferentiated and uniformly granular. The prodissoconch-II stage which develops during the next two weeks acquires a columnar organization. Metamorphosis is characterized by the formation of the dissoconch shell with all the elements of an adult shell and marks the onset of the development of the nacre. Growth of the black-lip pearl oyster, Pinctada margaritifera, at nine culture sites of French Polynesia: synthesis of several sampling designs conducted between 1994 and 1999 ArchiMer

Between 1994 and 1999, several sampling designs were used to measure the growth of the black-lip pearl oyster, Pinctada margaritifera var. cumingi at different sites in French Polynesia. Using a common statistical method, growth data were analysed for nine sites and showed significant geographic variability. Parameters of the Von Bertalanffy model, fitted on the nine data sets, ranged from 147 to 186.5 mm for the H-infinity parameter and from 0.42 to 0.58 year(-1) for the k parameter. For cultured pearl production, a mantle allograft from a donor is implanted into the gonad of a recipient oyster, together with a small inorganic bead. Because of the dedifferentiation of cells during the first steps of the host oyster's immunological reaction, so far the fate of the graft and its exact role in the process of pearl formation could not be determined via classical histological methods. Genetic structure at different spatial scales in the pearl oyster (Pinctada margaritifera cumingii) in French Polynesian lagoons: beware of sampling strategy and genetic patchiness ArchiMer

In order to study further the genetic structure of the pearl oyster Pinctada margaritifera in French Polynesia with a special consideration for the sampling scale, we analyzed or re-analyzed sets of data based on nuclear DNA markers obtained at different spatial scales. At a large scale (several 1,000 km), the remote Marquesas Islands were confirmed to be significantly differentiated from Tuamotu-Gambier and Society archipelagos, with a marked difference however for the two main islands that are different from each other. Effect of food conditioning on gonadic activity in the oyster Pinctada margaritifera ArchiMer

Preliminary experiments allowed defining basic requirements of Pinctada margaritifera for somatic and gonadic growth in controlled conditions. This study was continued by observation of the gametogenesis at a cellular level. In April/May 2008, 125 oysters were conditioned for 30 days in 25 tubular tanks (30 L). Water was renewed 4 times/hour. A mixed (v:v) diet of Isochrysis galbana (T-Iso) and Chaetoceros gracilis was supplied continuously. Although this technique has been used for many years in French Polynesia, the grafting process is still not standardised. Determination of gender in the pearl oyster pinctada margaritifera ArchiMer

Hatchery-scale trials using cryopreserved spermatozoa of black-lip pearl oyster, Pinctada margaritifera ArchiMer

Cryopreservation is a valuable tool for genetic improvement programs. Several bivalve mollusc species have already been the subject of such programs and the Tahitian black pearl oyster industry is now planning the development of selective breeding for desirable traits in Pinctada margaritifera. The ability to cryopreserve spermatozoa would, therefore, offer significant benefits to the cultured black pearl industry. Spermatozoa were cryopreserved with CPA 0.7 M trehalose in 0.8 M Me2SO and a two-step freezing process was used: straws were first maintained in nitrogen vapour for 10 minutes, then directly plunged into liquid nitrogen and stored for one week before use.
